Mechanism:Converted to an active metabolite. Inhibits DNA synthesis as also that of RNA and protein. May be selectively toxic to hypoxic cells because it may generate superoxide and hydroxylradicals. Effects are most marked in the late Gl and early S phases of cell cycle.
Indications:Ademocarcinoma, lymphosarcoma and seminoma. Superficial bladder cancer as a adjuvant in pterygia. Contralndications: : Haemorrhagic tendencies, bone marrow depression, not to be given I.M. or S.C., thrombocytopenia.
Adverse effects and Precautions As for Doxorubicin
Dosage:Adults : 6-10 mg I.V. twice weekly or 0.05mg / kg body weight daily for 5 days repeating schedule after 2 days to a total Dose of 60-100 mg.
